Current DNA ligation methods often require harsh conditions or external reagents, limiting their applicability in biological settings. Here, the authors introduce a DNA chemical ligation technique that involves the reaction between 3’-phosphorothioate DNA and 5’-dinitrobenzenesulfonamide DNA to yield a P3’ → N5’ phosphoramidate linkage, achieving over 80% yield at 37 °C and pH 8 under near-physiological conditions without the need for external reagents.
