Prefrontal cortex AMPA receptor plasticity is crucial for cue-induced relapse to heroin-seeking

This study demonstrates that relapse in heroin-addicted rats requires endocytosis of the AMPA receptor subunit GluR2 in the medial prefrontal cortex. When endocytosis was inhibited in this specific brain structure, the addicted, but abstinent, rats were less intent on seeking heroin when re-exposed to drug-associated cues.

Lasting synaptic changes underlie attention deficits caused by nicotine exposure during adolescence

The authors report that adolescent nicotine exposure in rats induces lasting attentional disturbances and reduces mGluR2 function in the PFC. Restoring mGluR2 activity rescued attention specifically.

Shisa6 traps AMPA receptors at postsynaptic sites and prevents their desensitization during synaptic activity

Auxiliary AMPA receptor subunits can affect gating and surface mobility. Here the authors show that Shisa6 traps AMPA receptors at postsynaptic sites via PSD-95, and keeps them in an activated state in the presence of glutamate, preventing full desensitization and consequently synaptic depression.

Retrieval-specific endocytosis of GluA2-AMPARs underlies adaptive reconsolidation of contextual fear

The authors show that retrieval of fear memory modifies the membrane expression of GluA2-containing AMPA receptors and synaptic strength in the dorsal hippocampus. This synaptic plasticity exerts an inhibitory constraint on memory strengthening and underlies the loss of fear response by reinterpretation of memory content during adaptive reconsolidation.
