Using more than 100 independent iPSC lines derived from patients with Alzheimer’s disease, the authors discovered loci associated with the neuronal production of amyloid β and confirmed their influence using RNA interference.

Discovery of widespread transcription initiation at microsatellites predictable by sequence-based deep neural network

Mammalian genomes are scattered with repetitive sequences, but their biology remains largely elusive. Here, the authors show that transcription can initiate from short tandem repetitive sequences, and that genetic variants linked to human diseases are preferentially found at repeats with high transcription initiation level.

A coding variant in RARG confers susceptibility to anthracycline-induced cardiotoxicity in childhood cancer

Colin Ross and colleagues report the results of a genome-wide association study of anthracycline-induced cardiotoxicity in children treated for cancer. They identify a nonsynonymous coding variant in RARG associated with roughly fivefold higher risk of developing this severe adverse drug reaction.

Dmrt2 promotes transition of endochondral bone formation by linking Sox9 and Runx2

Koichiro Ono et al. report that Dmrt2 is a critical transcription factor for chondrocyte maturation during endochondral ossification. The study shows that Dmrt2 is the direct target of Sox9 and acts as a transcriptional coactivator of Runx2 providing novel insights into the transcription factor network during skeletal development.
