Endoplasmic reticulum (ER)-stress signaling shapes tumor immunity, yet the underlying molecular mechanisms remain poorly defined. Here, the authors show that the glycosylation enzyme DPM1 regulates the activity of ER-stress sensor IRE1, and that low DPM1 expression is associated with improved responses to immunotherapy in patients with colorectal cancer.
