S100A4, a calcium-binding protein generally associated with differentiation, is expressed by stromal cells in adipose tissue. This study shows that it protects mice from high-fat diet-induced obesity and glucose intolerance. S100A4 deficiency aggravates hepatic lipid accumulation and inflammation, and both extracellular and endogenous S100A4 promotes insulin signaling activation. S100A4 may therefore represent a potential candidate for the treatment of diet-induced obesity.
