Self-assembled nanogels from polysaccharides grafted with short oligonucleotides: control of size and thermoresponsiveness

The construction of self-assembled nanogels with oligonucleotide double strands as the association points was achieved by grafting short-chain (4-mer and 6-mer) oligonucleotides containing bridged nucleic acids (BNA), an artificial nucleic acid, onto polysaccharides. The oligonucleotide-grafted polysaccharides, composed of oligonucleotides with varying chain lengths, sequences, and BNA substitution numbers, formed nanogels in water, and their aggregation behavior was found to change with temperature.

Mucosal diseases are becoming more prevalent and needle-free vaccines could be instrumental in combating this. A nanometre-sized hydrogel consisting of a cationic type of cholesteryl group bearing pullulan has now been used as an intranasal vaccine-delivery system.

Enhanced HSP70 lysine methylation promotes proliferation of cancer cells through activation of Aurora kinase B

HSP70 is a molecular chaperone that aids protein folding. In this study, HSP70 is shown to be methylated and this post-translationally modified protein is elevated in expression in human cancers and promotes the activity of Aurora kinase B.

Polysaccharide nanogel–cyclodextrin system as an artificial chaperone for in vitro protein synthesis of green fluorescent protein

The molecular chaperone function of polysaccharide nanogel was demonstrated for the folding of newly synthesized green fluorescent protein (GFP) in a cell-free protein synthesis system. Nanogel comprising cholesteryl group-bearing pullulan (CHP) trapped unfolded or partially folded GFP expressed in the cell-free system. The structure of the CHP nanogel was disrupted by the addition of cyclodextrins, and the protein complexed with the nanogel was released and folded into the mature form.

Protein nanogelation using vitamin B₆-bearing pullulan: effect of zinc ions

Protein nanogels were prepared by using the vitamin B₆ (pyridoxal)-bearing pullulan (PLPP) as a bio-crosslinker. The binding of the PLPP to the serum albumin via the formation of Schiff bases was significantly enhanced in the presence of zinc ions because of the coordination of the Schiff base to metal ions. Dynamic light scattering measurements, high-performance liquid chromatography, and transmission electron microscopy confirmed that the zinc ions enhanced the ability of PLPP to form nanogels by crosslinking with the anionic BSA despite the strong electrostatic repulsion between the two molecules.

Nanoscale structures of radiation-grafted polymer electrolyte membranes investigated via a small-angle neutron scattering technique

The nanoscale structures of graft-type PEMs, prepared by radiation-induced graft polymerization of styrene onto poly(ethylene-co-tetrafluoroethylene) (ETFE) films followed by sulfonation, were investigated using a small-angle neutron scattering (SANS) technique. The SANS profiles of the grafted films showed shoulder peaks at a d-spacing of ∼30 nm, which were attributed to the polystyrene grafts introduced into the amorphous phases between the ETFE lamellar crystals. In the ETFE PEMs, this d-spacing increased to 34 nm because the graft regions were enlarged by the volume of the attached sulfonic acid groups.
