Prostaglandin E₂–EP4 signaling promotes immune inflammation through T_H1 cell differentiation and T_H17 cell expansion

Prostaglandins play a key role in inflammation in a variety of settings. Now, Shuh Narumiya and colleagues show that prostaglandin E2 drives the production of inflammatory T helper cells, and that this can be blocked by inhibiting its EP4 receptor subtype. EP4 inhibitors were also effective at inhibiting disease pathogenesis in animal models of two inflammatory diseases.

Prostaglandin F_2α receptor signaling facilitates bleomycin-induced pulmonary fibrosis independently of transforming growth factor-β

Idiopathic pulmonary fibrosis, or lung scarring, is known, at least in part, to be driven by TGF-β signaling. Shuh Narumiya and colleagues now find that prostaglandin F_2α receptor also has a key role in this disease, independently of TGF-β signaling, and that its genetic deletion ameliorates disease progression in a mouse model.

Mechanisms of prostaglandin E2 (PGE2)-mediated immunosuppression in the tumor microenvironment (TME) have been previously reported. Here, the authors profile PGE2 functions in human cancer, suggesting that prostaglandin E2-mediated signaling impairs the activity of human CD8+ T cells and macrophages by altering bioenergetics and ribosome biogenesis.

Receptor-activator of NF-κB ligand (RANKL) and its receptor RANK are known to be essential regulators of bone remodelling, lymph node organogenesis and formation of a lactating mammary gland, but the functional relevance of their expression in the brain has been unclear. RANKL and RANK are now reported to have an essential role in the brain, with the central injection of RANKL into mice and rats triggering severe fever, and a further potential role in the control of thermoregulation in females.

Lipocalin-type prostaglandin D synthase regulates light-induced phase advance of the central circadian rhythm in mice

Kawaguchi et al. show that mice deficient in lipocalin-type prostaglandin (PG) D synthase (L-PGDS) exhibit impaired light-induced phase advance, but normal phase delay and nonvisual light responses. This study suggests the role of L-PGDS for the light-induced phase advance possibly via a chemoattractant receptor DP2/CRTH2.
