Cofilin-1 regulates cytoskeletal dynamics, but its role in mitochondria is less clear. Here, the authors demonstrate that N-terminal SUMOylation directs cofilin-1 to mitochondria, where it triggers mitochondrial dysfunction and aberrant apoptotic pathways.
