Through single molecule imaging Yoshioka et al reveal a positive feedback loop between PI(4,5)P2 and PTEN to promote PTEN membrane binding in vitro. They also show that the PI(4,5)P2 binding domain is critical for PTEN distribution and migration in Dictyostelium cells. This work illustrates the power of single molecule imaging in probing protein-lipid interactions.
