Structural basis of protein arginine rhamnosylation by glycosyltransferase EarP

The crystal structure of EarP, an inverting glycosyltransferase that generates rhamnosyl-arginine modifications, suggests that the enzyme uses an S_N2 reaction mechanism that may involve perturbation of the donor sugar nucleotide conformation.

Structural basis of the regulation of the normal and oncogenic methylation of nucleosomal histone H3 Lys36 by NSD2

An upregulation of NSD2, a histone H3 lysine 36 (H3K36) methyltransferase is linked to multiple myeloma and other types of cancer. Here, the authors provide insights into the regulatory mechanism of NSD2 by determining the 2.8 Å cryo-EM structure of the NSD2 bound nucleosome complex, and based on MD simulations they discuss how two oncogenic mutations enhance NSD2 activity.

Genetic and functional diversity of β-N-acetylgalactosamine-targeting glycosidases expanded by deep-sea metagenome analysis

Four β-N-acetylgalactosaminidase gene families were identified via deep-sea metagenome analysis. The biochemical and structural characterization of the aforementioned enzymes revealed their functional diversity and monophyletic evolutionary history.

Structure of the human Bre1 complex bound to the nucleosome

The structure of the nucleosome-bound human Bre1 complex reveals that its two RING domains bind the acidic patch and nucleosomal DNA, directing the E2 enzyme and ubiquitin for H2BK120-specific ubiquitination. The binding mode suggests a possible regulatory mechanism through nucleosomal DNA flexibility.

LimF is a versatile prenyltransferase for histidine-C-geranylation on diverse non-natural substrates

His-C2-directed modification is chemically challenging and rarely occurs in nature due to the low reactivity of this position. Now, the prenlytransferase LimF has been discovered and applied for geranylation of histidine-containing peptides and imidazole-containing small molecules, showcasing the versatility of this biocatalyst.

Ribosomal synthesis and de novo discovery of bioactive foldamer peptides containing cyclic β-amino acids

Cyclic β-amino acids can add useful properties to peptides, such as inducing turn structures or providing resistance to proteases. To harness these properties up to ten consecutive cyclic β-amino acids have now been ribosomally incorporated via genetic code reprogramming into a foldamer peptide library that has been screened for potent binders against a protein target, human factor XIIa.
