Iron homeostasis and cell clonality drive cancer-associated intestinal DNA methylation drift in aging

Krepelova and colleagues identify an aging- and colon cancer-associated DNA methylation drift in healthy and cancerous human colon samples, echoed in the mouse intestinal epithelium. They show it expands via intestinal cell clonality and is driven by dysregulated iron metabolism.

Most regions of the mammalian genome replicate both alleles in a synchronous manner, but some loci have been found to replicate asynchronously and the time of replication of each allele is different. Here the authors, by employing clonal mouse cells from a hybrid strain chart replication timing over the entire genome, using polymorphisms to distinguish between the paternal and maternal alleles.

TET2 and TET3 loss disrupts small intestine differentiation and homeostasis

DNA demethylation is known to be critical for the development and function of many tissues. Here the authors show that it is also required for intestinal lineage differentiation, and that mice lacking DNA demethylases have altered microbiomes and a predisposition to inflammation.

The microbiota programs DNA methylation to control intestinal homeostasis and inflammation

A comparison of DNA methylation patterns between conventionally raised and germ-free mice shows that the presence of a commensal microbiota induces methylation changes at regulatory elements in a subset of genes that are important for intestinal homeostasis.

B cell development involves sequential rearrangement of the immunoglobulin chains, but fine control over the selection process remains a mystery. Here the authors show that individual alleles in pre-B cells are clonally unique and result from stochastic activation of V gene segments to induce optimal generation of a diverse repertoire.

Clonal allelic predetermination of immunoglobulin-κ rearrangement

Immunoglobulin genes are expressed from either the maternal or paternal chromosome; it is now shown that in early haematopoietic stem cells, an individual cell can choose either of the two alleles, but as they develop they become committed to only one.

De novo DNA methylation promoted by G9a prevents reprogramming of embryonically silenced genes

G9a is involved in gene silencing during early embryonic development, catalyzing the methylation of H3K9, which results in heterochromatinization, and also promoting methylation of DNA de novo. These two G9a activities are now dissected, and de novo DNA methylation is shown to occur via recruitment of Dnmt3a/3b and to be necessary and sufficient to prevent reprogramming.
