The exchanger operates in forward mode, transporting cytosolic Na+ into the matrix in exchange for matrix Ca2+, which has entered through the MCU. Mitofusin 2 is located in the OMM and functionally detached from the exchanger. After mitochondrial depolarisation (right-hand panel), the exchanger is now functionally coupled to mitofusin 2 and can operate in both forward and reverse modes.
