In WT animals, siRNAs undergo amplification within the M compartment through pUG RNA-siRNA cycles. These siRNAs can degrade RNA in the cytoplasm when bound to WAGOs. Additionally, pUG RNAs can be memorized by ZNFX-1 within the Z compartment, facilitating TEI. Upon depletion of HERD-1, the PZM proteins interaction network becomes mixed, bringing previously separated small pathways into proximity. This coalescence may lead to increased incorporation of pUG RNAs or proteins into TEI and reduced distribution of siRNAs or proteins to the cytoplasm for RNAi-mediated RNA degradation.
