9-week-old male and female C57BL/6 J mice placed on STC were administered with AAV encoding Gpr110 (AAV-GPR110) or Gfp (AAV-GFP) driven by thyroxine-binding globulin (Tbg) promoter via tail-vein injection (2×1011 copies of AAV/mouse). After 1 week, the mice were switched to CDAHF60 diet for 12 weeks (n = 6/group). a,c, AAV-mediated selective overexpression of Gpr110 in the liver, but not in subcutaneous white adipose tissue (sWAT), epididymal white adipose tissue (eWAT), gastrocnemius muscle (Gastroc) or hypothalamus (Hypo) in male (a) and female (c) mice was confirmed by quantitative real-time PCR analysis. b,d, Hepatic protein expression of GPR110 induced by AAV-GPR110 in male (b) and female (d) mice was validated by western blot.
