(a) Representative immunoblots for CBP and GCN5 histone acetyl-transferases in young and aged MSCs. β-actin, loading control. n = 2 biologically independent replicates. (b) Basal (top) and maximal (bottom) Oxygen Consumption Rates (OCR) in young and aged MSCs, treated with or without 100 μM Etomoxir for 1 hour, prior to the assay. Measurements were made using the SeaHorse XF24 extracellular flux analyzer instrument. n = 3 biologically independent replicates. (c) Basal (top) and maximal (bottom) OCR in young and aged MSCs. Cells were incubated overnight with substrate-limited medium. On the day of the assay, BSA-Palmitate was injected to cells. Measurements were made using the SeaHorse XF24 extracellular flux analyzer instrument. n = 4 biologically independent replicates in young and n = 3 biologically independent replicates in old. Data in (b) and (c) are presented as mean ± SEM. Statistical significance was determined using two-sided unpaired t-Test.
