Whole-brain tissue mapping toolkit using large-scale highly multiplexed immunofluorescence imaging and deep neural networks

It is challenging to map complex processes in brain tissue. Here the authors report a toolkit enabling large-scale multiplexed IHC and automated cell classification whereby they use a conventional epifluorescence microscope and deep neural networks to phenotype all major cell classes of the brain.

A role for interleukin-2 trans-presentation in dendritic cell–mediated T cell activation in humans, as revealed by daclizumab therapy

Dendritic cells produce interleukin-2 (IL-2) and express the IL-2 receptor subunit CD25. Bibiana Bielekova and her colleagues show that dendritic cells, upon interacting with cognate T cells, secrete IL-2 into the immune synapse and use their CD25 to trans-present IL-2 to T cells, facilitating early IL-2 signaling in T cells. Inhibition of CD25 by the monoclonal antibody daclizumab prevents T cell activation and may partly account for the therapeutic effects of daclizumab in patients with multiple sclerosis.

Differentiation of human and murine induced pluripotent stem cells to microglia-like cells

Pandya et al. describe a protocol to differentiate human and mouse iPSCs into cells with the phenotype, transcriptional profile and functional properties of microglia. The treatment of murine intracranial malignant gliomas with these cells demonstrates their potential clinical use. These microglia-like cells will enable further studies into the role of microglia in health and disease.

Murray et al. present a multiplexed fluorescence-based immunohistochemistry approach and use it to screen up to 100 protein markers in the human olfactory bulb from neurologically healthy, Alzheimer’s, and Parkinson’s disease patients. In doing so, they identify differentially expressed biomarkers in Alzheimer’s, and Parkinson’s disease.

Nikorn Pothayee et al. show that early neural precursor cells (NPCs) derived from the embryonic telencephalon or midbrain can develop into brain-like tissue when implanted into the rat brain ventricle. Telencephalon-derived NPCs also form brain tissue in the host cortex when implanted into a CSF-filled cavity formed by cortical ablation.
