Oxygen transposition of formamide to α-aminoketone moiety in a carbene-initiated domino reaction

C=O cleavage of formamides and full transfer of the resulting fragments into final products are highly appealing, particularly in multi-component reactions for the synthesis of structurally complex molecules. Now three-component reactions of diazo compounds with allylic substrates and formamides are presented, giving extensively reorganized products via multi-step relay.

Heterozygous deletion of chromosome 17p renders prostate cancer vulnerable to inhibition of RNA polymerase II

Heterozygous loss of chromosome 17p is a common genomic event in castration-resistant prostate cancers (CRPC). Here, the authors use CRISPR-based screen to identify RBX1 as a target gene for CRPC with 17p loss and targeting RBX1 sensitizes CRPC to the treatment of α-amanitin antibody conjugates.

Mutually exclusive acetylation and ubiquitylation of the splicing factor SRSF5 control tumor growth

Changes in glucose metabolism can lead to tumor development, but the involvement of splicing factors is unclear. Here, the authors screened for SR proteins and identified SRSF5 stability is enhanced in response to glucose elevation to promote alternative splicing of CCAR1 which facilitates tumor growth.
