Here, the authors investigate whether antibodies specific to the antigen peptide-MHC class II complex are generated during the immune response and whether these antibodies can limit hyperimmune responses. They identify a class of antibodies, called iTabs, that recognize peptide-MHC class II complexes and block TCR engagement, ultimately limiting antigen-specific CD4 + T cell responses, and provide Cryo-EM structures detailing the iTab-mediated recognition of the antigen.
