Angiogenesis induced by CNS inflammation promotes neuronal remodeling through vessel-derived prostacyclin

Angiogenesis is induced in response to central nervous system (CNS) injury and inflammation. Toshihide Yamashita and colleagues show that in a mouse model of multiple sclerosis, new vessels form around inflammatory lesions and promote neuronal remodeling, rewiring of the corticospinal tract, and recovery of motor function in these mice. Prostacyclin is released from these vessels and promotes neuronal outgrowth by signaling through the IP receptor on neurons.

Layer V cortical neurons require microglial support for survival during postnatal development

In this study, the authors show that layer V projection neurons require the presence of subcortical CX3CR1-positive microglia for survival. IGF1 secretion from these microglia appears to be necessary for this trophic effect. Inhibition of the microglial cell activation abrogates IGF1 secretion and compromises neuronal survival.

Uterine adenomyosis often co-occurs with endometriosis or leiomyoma, but little is known about its molecular underpinnings. Wnt5a-Ror2 signaling between osteoblast-lineage cells and osteoclast precursors enhances osteoclastogenesis

In a new study, Yasuhiro Kobayashi and his colleagues show that noncanonical Wnt signaling regulates balanced osteoblast-induced osteoclastogenesis during normal physiology and that this pathway is perturbed in pathophysiological states, such as rheumatoid arthritis. These results explain further how osteoblasts cross-talk with preosteoclasts to ensure matched bone resorption with bone formation during skeletal homeostasis in the adult and also suggest a new target to treat arthritis.

miR-124a is required for hippocampal axogenesis and retinal cone survival through Lhx2 suppression

The authors investigate the in vivo function of microRNA-124a by specifically disrupting Rncr3, the dominant source of miR-124a in mice. Their results suggest that miR-124a is important for maturation and survival of dentate gyrus neurons and retinal cones and acts by repressing Lhx2 translation.

Hiroyuki Mano and colleagues report fusions involving DUX4 in 16.4% of Ph-negative adolescent and young adult acute lymphoblastic leukemia (AYA-ALL) cases. Transplantation assays in mice support an oncogenic role for the DUX4-IGH fusion gene, which expresses DUX4 protein with an aberrant C terminus at high levels in patients with AYA-ALL.

Given that archival FFPE tissue is of great interest for genomic analysis, but difficult to reliably analyze, this tool may improve the ability of researchers to probe sequencing data from these samples.
