Single crystals of mechanically entwined helical covalent polymers

Single crystals of a helical covalent polymer have been obtained from an achiral monomer through spiroborate formation. Polymerization and crystallization occur simultaneously to give a network of pairs of entwined helical strands of the same handedness. No strong non-covalent interactions were observed between the two helical polymers forming a pair; instead, each interacts with neighbouring pairs through hydrogen bonding.

Targeting Smurf1 to block PDK1–Akt signaling in KRAS-mutated colorectal cancer

SMAD ubiquitylation regulatory factor 1 (Smurf1) was identified as a critical activator of the phosphoinositide 3-kinase (PI3K)–Akt pathway by promoting phosphoinositide-dependent protein kinase 1 (PDK1) neddylation and cytoplasmic complex of PDK1 assembled with Smurf1 and SETDB1 (cCOMPASS) assembly. Smurf1-antagonizing repressor of tumor 1 (SMART1), a highly selective degrader for Smurf1, was developed and suppressed the tumor growth of Kirsten rat sarcoma viral oncogene homolog (KRAS) mutant colorectal cancer.

The synthesis of graphynes has often been limited to using irreversible coupling reactions that are likely to result in materials that lack long-range order. Now, a periodically sp–sp ²-hybridized carbon allotrope, γ-graphyne, is prepared using a reversible dynamic alkyne metathesis and managing the balance between kinetic and thermodynamic control. This material’s interlayer stacking and folding behaviour are also revealed.

Alkyl and aryl polycyanurate networks have now been prepared through polymerization of diols and substituted triazines via a dynamic S_NAr reaction. When treated with excess mono alcohol or phenol, the polycyanurate networks can be depolymerized into the starting monomers, which can be separated and reused, thus achieving closed-loop recycling.
