TMexCD1-TOprJ1 is the paradigm for transferable tigecycline resistance by efflux pumps. Here, authors report biochemical and cryo-EM structural description of TMexCD1-TOprJ1 tripartite machinery, illuminating its assembly and inhibition mechanism. It benefits developing next-generation of efflux pump-targeted antimicrobials.
