A multifaceted strategy for intra- and extracellular nucleic acid regulation to alleviate intervertebral disc degeneration

Abnormal accumulation of intra- and extracellular free nucleic acids contributes to the chronic inflammation driving intervertebral disc degeneration (IVDD). Here, the authors report a nanovesicle system to inhibit mitochondrial DNA release in nucleus pulposus cells and remove extracellular DNA from the disc microenvironment to block inflammation for treatment of IVDD.

DDX5 inhibits hyaline cartilage fibrosis and degradation in osteoarthritis via alternative splicing and G-quadruplex unwinding

Liu et al. identify downregulation of DDX5, an RNA helicase, in the cartilage of patients and mouse model of osteoarthritis. Targeted upregulation of DDX5 in mouse chondrocytes inhibits hyaline cartilage fibrosis and degradation via pre-mRNA splicing and G4 unwinding, a potential therapeutic strategy against osteoarthritis.
