Anti-Inflammatory Applications of Forsythia Suspensa Extracts
Summary
Forsythia suspensa, known in traditional Chinese medicine as Forsythiae Fructus, has been employed for centuries as a heat‐clearing and detoxifying agent. Modern pharmacological investigations have isolated key bioactive constituents—including phenylethanoid glycosides (forsythiaside A and B), lignans (arctiin, phillyrin) and flavonoids—that exhibit potent anti‐inflammatory effects. These extracts are capable of inhibiting cyclooxygenase-2 expression and suppressing nuclear factor-κB activation, thereby reducing the production of nitric oxide and pro‐inflammatory cytokines such as interleukin-1β, interleukin-6 and tumour necrosis factor-α. Both in vitro and in vivo models show attenuation of neutrophil infiltration, oxidative stress and tissue damage. Advances in metabolomic and proteomic profiling have unveiled multi‐target mechanisms, demonstrating effects on nicotinate and nicotinamide metabolism, purine and pyrimidine pathways, and modulation of extracellular matrix proteins. Together, these findings underscore the potential of Forsythia suspensa extracts as adjunctive therapies in chronic inflammatory conditions, with applications ranging from dermatological to neuroinflammatory disorders. Ongoing efforts focus on optimising extraction methods, enhancing bioavailability and establishing safety profiles to support clinical translation.
