Avocado/Soybean Unsaponifiables in Osteoarthritis Management

Summary

Avocado/soybean unsaponifiables (ASU) are a defined mixture of lipid extracts derived from avocado and soybean oils. These extracts are rich in plant sterols, tocopherols and phytosterols, which collectively exert anti-inflammatory and pro-anabolic effects on joint tissues. In osteoarthritis, ASU has been shown to inhibit key catabolic enzymes and inflammatory mediators—such as matrix metalloproteinases and prostaglandin E₂—while enhancing synthesis of cartilage matrix components including glycosaminoglycans and collagen. Clinically, ASU is employed as a slow-acting symptomatic therapy for hip and knee osteoarthritis, providing pain relief and functional improvement with an excellent safety profile. Beyond symptom control, preclinical evidence suggests potential disease-modifying properties through suppression of nitric oxide synthase activity and modulation of cytokine networks. ASU formulations are authorised in numerous countries as adjuncts to standard care, and ongoing research seeks to define optimal dosing, active constituents and combination strategies with other nutraceuticals or pharmaceutical agents.
