Summary

Calcineurin is a ubiquitous serine/threonine phosphatase that decodes calcium signals to regulate an array of cellular functions. Its activation requires calcium binding to calmodulin and the cooperative displacement of an autoinhibitory domain, allowing catalytic access to phosphorylated substrates. Calcineurin recognises targets via conserved docking motifs, notably PxIxIT and LxVP sequences, and is subject to post-translational regulation such as palmitoylation, which confers unique membrane localisation. This palmitoylation-dependent targeting enables calcineurin to associate with the phosphatidylinositol 4-kinase complex, where dephosphorylation enhances lipid kinase activity and modulates phosphoinositide production in receptor signalling. Another work elucidated the molecular determinants of substrate specificity by demonstrating how a cellular exchanger is selectively dephosphorylated via combined recognition motifs and dynamic charge interactions, revealing how calcineurin discriminates among phosphorylation sites and recruits distinct targets to regulate pH homeostasis.
