Canine Parvovirus Epidemiology and Diagnostics
Summary
Canine parvovirus (CPV) is a highly contagious pathogen that emerged in the late 1970s and rapidly achieved global distribution among domestic and wild canids. Transmission occurs primarily by the faecal–oral route, facilitated by the remarkable environmental stability of the non-enveloped, single-stranded DNA virus. Young and unvaccinated dogs are most susceptible, often presenting with severe haemorrhagic enteritis, dehydration and high mortality rates. Three antigenic variants (2a, 2b and 2c) continue to co-circulate, displaying regional predominance—2a in parts of Asia, 2c in Europe and Latin America and 2b in other areas—with ongoing mutation of the capsid VP2 protein influencing host range, immune evasion and vaccine efficacy. Robust surveillance relies on a combination of point-of-care antigen tests, serological assays, molecular diagnostics and genome sequencing. Seroprevalence studies gauge population immunity, while advanced molecular methods, including real-time PCR and deep-sequencing protocols, resolve mixed infections, co-circulation of variants and recombinant strains. These diagnostic advances underpin timely outbreak control and inform refined vaccination strategies, contributing to a global effort to monitor viral evolution and reduce canine morbidity and mortality.

Recent structural analyses have applied high-resolution cryo-electron microscopy to map the canine antibody response to live CPV vaccination. By isolating polyclonal Fab fragments from immunised dogs at eight and twelve weeks post-vaccination, researchers reconstructed multiple Fab–virus complexes using subparticle extraction and correlated classification. Five distinct antibody binding sites were identified, each overlapping the transferrin receptor attachment region on the VP2 capsid. This work reveals a limited, convergent immune fingerprint elicited by current vaccines and highlights precise epitope targets that could be harnessed to improve diagnostic reagent design, monitor vaccine-driven antigenic drift and guide next-generation vaccine formulations.
