An integrative genomic and transcriptomic study of spontaneous canine soft tissue sarcomas characterised fibrosarcoma, pleomorphic sarcoma and peripheral nerve sheath tumours through whole-exome sequencing, RNA-seq and immunohistochemistry. This work identified recurrent mutations in TP53 and chromatin-modifying genes (notably KMT2D) in over one-third of cases, and uncovered PDGFB fusion transcripts predicting sensitivity to PDGFR inhibitors. Transcriptomic clustering delineated four molecular subgroups with distinct extracellular matrix, homeobox transcription factor or immune-infiltration signatures, providing a framework for refined diagnosis, comparative oncology studies and prioritisation of targeted therapy trials in pet dogs.
