By integrating target-specific ligation and cell-free translation, this approach delivers sensitive detection of multiple respiratory pathogens without thermal control or complex equipment. In parallel, an automated computational platform has expanded the CFPS sensor toolkit by converting protein-binding RNA aptamers into riboswitches. Engineered sensors exhibit up to 16-fold translational regulation in response to human biomarkers, demonstrating low-cost, cell-free assays for point-of-care protein detection and illustrating how in silico design enhances riboswitch accuracy.
