Summary

Second-generation biofuels derive from lignocellulosic biomass, in which cellulose is hydrolysed to cellobiose, a disaccharide of two glucose units. Efficient fermentation of cellobiose is essential to lower production costs and improve yields. Early approaches relied on exogenous cellulases to generate glucose for conventional yeast fermentation. More recent strategies focus on microbial consolidation of saccharification and fermentation in a single organism or consortium, known as consolidated bioprocessing, thereby reducing enzyme requirements and process steps. Engineering of Saccharomyces cerevisiae and thermotolerant yeasts such as Ogataea polymorpha has introduced heterologous cellodextrin transporters and β-glucosidases or cellobiose phosphorylases, enabling direct uptake and intracellular hydrolysis of cellobiose. Metabolic engineering efforts have targeted transporter expression, enzyme activities, regulatory networks and transcription factors to optimise pathway flux, manage ATP homoeostasis and mitigate inhibitory effects in mixed-sugar streams. Co-cultivation and kinetic modelling approaches further aim to balance specialist strains for parallel fermentation of cellobiose and pentoses such as xylose. Collectively, these bioengineering and process-integration advances have enhanced ethanol productivity and yield, moving closer to economically viable large-scale biofuel production from plant biomass.
