Chitinase-Mediated Defense Mechanisms in Transgenic Plants
Summary
Chitinases are hydrolytic enzymes that target chitin, a structural polysaccharide in fungal cell walls and insect exoskeletons. In plants, these enzymes form part of the pathogenesis‐related protein repertoire and contribute to basal immunity by degrading invading pathogen cell walls and releasing elicitor fragments. Transgenic approaches harness these activities by introducing or overexpressing chitinase genes from diverse sources—plant, microbial or pathogen—to confer enhanced disease resistance. Transgenic plants often display elevated levels of reactive oxygen species bursts, activation of mitogen‐activated protein kinase pathways and upregulation of defence‐related genes, culminating in broad‐spectrum resistance against fungi, oomycetes and certain bacteria. Chitinase expression can be driven by constitutive or pathogen‐inducible promoters, enabling finely tuned defence responses. Beyond direct antimicrobial action, chitinase transgenes can prime the host immune system, leading to faster and stronger activation upon subsequent pathogen challenge. Field trials in key crops such as rice, wheat and tomato have demonstrated reduced disease incidence and improved yield stability under pathogen pressure. Ongoing efforts focus on stacking multiple chitinase classes, optimising subcellular targeting and combining chitinase transgenes with other defence genes to achieve durable resistance while minimising off‐target effects on non‐pathogenic symbionts.
