Summary
Conjugated linoleic acid (CLA) refers to a family of positional and geometric isomers of linoleic acid found predominantly in ruminant-derived foods. Two principal isomers, c9,t11 and t10,c12 CLA, exhibit distinct biological activities. Mechanistically, CLA modulates lipid metabolism through activation of nuclear receptors such as peroxisome proliferator-activated receptors (PPARs), influences energy homeostasis by promoting mitochondrial uncoupling and adipose tissue browning, and alters secretion of bioactive peptides from adipocytes. Preclinical models consistently demonstrate anti-adipogenic and anti-atherogenic properties, while human trials have reported modest reductions in body fat alongside variable effects on insulin sensitivity, hepatic lipid accumulation and markers of inflammation. The duality of CLA’s actions—ranging from beneficial immunomodulation and lipid-lowering to potential induction of insulin resistance and hepatic steatosis—underscores the need for isomer-specific evaluation. As obesity and related metabolic disorders rise globally, a nuanced understanding of CLA’s health impacts and delivery strategies is essential for its safe incorporation into dietary and therapeutic programmes.
