Cripto-1 Signaling in Cancer and Stem Cell Biology

Summary

Cripto-1 is a GPI-anchored oncofetal protein of the EGF-CFC family that functions as a co-receptor for transforming growth factor-β (TGF-β) ligands, notably Nodal. During embryogenesis, it orchestrates cell fate decisions, axis formation and stem cell maintenance via modulation of Smad, Wnt/β-catenin and Notch pathways. In adult tissues its expression is generally low but is reactivated in diverse cancers, where it drives cell proliferation, survival, motility and angiogenesis. Cripto-1 facilitates the epithelial–mesenchymal transition (EMT) underlying metastatic spread and sustains a subpopulation of cancer stem cells (CSCs) endowed with self-renewal and tumour-initiating capacity. By acting both as a membrane-bound co-receptor and a soluble ligand, it mediates autocrine and paracrine signalling that remodels the tumour microenvironment, fosters therapy resistance and is associated with adverse clinical outcomes. The centrality of Cripto-1 at the nexus of developmental and oncogenic networks underscores its promise as a biomarker and therapeutic target.
