Summary
Essential oils, complex mixtures of volatile phytochemicals predominantly comprising monoterpenes and sesquiterpenes, have garnered significant attention for their multifaceted roles in human health. These oils exert antimicrobial, antioxidant, anti-inflammatory and anticancer effects through mechanisms such as membrane disruption, modulation of oxidative stress and induction of programmed cell death. Advances in formulation science—particularly nano-delivery systems—have enhanced the bioavailability and targeted action of key constituents like citral and eugenol. Globally, essential oils are being integrated into complementary therapies, pharmaceutical preparations and hygienic applications to address drug resistance, chronic inflammation and tumour progression, underscoring their potential as multifunctional agents in both preventive and therapeutic contexts.

Innovations in nanostructured delivery have markedly improved the anticancer performance of oil-derived compounds. One seminal study demonstrated that incorporation of citral into lipid-based nanoparticles significantly increased its solubility, promoted selective uptake by breast cancer cells and induced cell cycle arrest at G2/M phase. This formulation triggered mitochondrial apoptosis and inhibited metastatic behaviour in vitro, while effectively reducing tumour burden in murine models without observable systemic toxicity. Earlier foundational work elucidated dual mechanisms of citral action: a p53-dependent pathway leading to apoptosis in p53-proficient cells, and an endoplasmic reticulum stress response in p53-deficient lines. Reactive oxygen species generation was identified as a proximal trigger, with downstream activation of stress markers and caspase cascades, thereby providing mechanistic insight for further therapeutic development.
