Hepatoprotective Natural Products and Phytotherapy
Summary
The liver is central to metabolic homeostasis, detoxification and the modulation of immune responses. Hepatoprotective natural products exploit diverse phytochemicals—such as flavonoids, terpenoids, alkaloids and polyphenols—to attenuate oxidative stress, inflammation and cell death pathways that underlie hepatic injury. Mechanistic studies reveal that these compounds often act by scavenging reactive oxygen species, upregulating endogenous antioxidant defences (for example glutathione and superoxide dismutase), stabilising cellular membranes and inhibiting pro-inflammatory mediators. Phytotherapy—the medicinal use of plant extracts—has advanced through the characterisation of active constituents and the optimisation of delivery platforms, including nano-encapsulation, to enhance bioavailability and target specificity. In preclinical models, agents such as silymarin, curcumin derivatives and quercetin glycosides demonstrate dose-dependent restoration of liver enzyme profiles (ALT, AST, ALP), histological architecture and bile flow. Emerging research integrates pharmacogenomics to tailor interventions according to genetic predisposition to liver disease. Collectively, these developments underscore the promise of natural products as adjuncts or alternatives to conventional hepatoprotective drugs, with potential to ameliorate toxin-induced, viral and metabolic liver disorders on a global scale.
