Hydrogel Technologies for Diabetic Wound Healing
Summary
Diabetic wounds pose a significant clinical challenge due to chronic inflammation, impaired angiogenesis and heightened infection risk. Hydrogels, with their high water content, biocompatibility and tunable mechanical properties, have emerged as versatile platforms for diabetic wound care. These networks of hydrophilic polymers can maintain a moist environment, support cell infiltration and serve as reservoirs for bioactive agents including antibiotics, growth factors, ions and extracellular vesicles. Recent advances have integrated stimuli-responsive elements that sense local pH, glucose or redox status to control release kinetics, as well as microneedle designs that enhance transdermal delivery. Immunomodulatory hydrogels now aim to convert pro-inflammatory macrophages into reparative phenotypes, while combined neurogenic and angiogenic cues accelerate tissue regeneration. Collectively, these hydrogel technologies promise to address the multifactorial barriers of diabetic wound healing and improve patient outcomes worldwide.
Research from Nature Portfolio
Recent studies have demonstrated a core–shell microneedle array capable of programmable reactive oxygen species generation under light stimulation to eradicate bacterial biofilms. Following initial debridement, the outer shell degrades to expose a core that neutralises inflammatory cytokines and releases an anti-fibrotic agent, thereby synchronising with the natural healing phases to reduce scarring and accelerate tissue repair in diabetic wound models.
In parallel, a photodynamic immunomodulatory microneedle bandage functionalised with hybrid nanoparticles has been devised to adapt to the diabetic biofilm microenvironment. By depleting glutathione, it enhances reactive species–mediated biofilm eradication and subsequently acts as a scavenger in low-glutathione regions to quell inflammation and promote macrophage polarisation towards the reparative M2 phenotype, resulting in improved wound closure and regeneration.
