Inositol Hexaphosphate Activity in Cancer Cell Proliferation

Summary

Inositol hexaphosphate, also known as phytate, is a naturally occurring polyphosphorylated carbohydrate abundant in whole grains, legumes and seeds. It has emerged as a multifaceted modulator of cancer cell proliferation, acting through direct and indirect mechanisms. InsP6 can arrest the cell cycle at key checkpoints, notably G1/S and G2/M, by upregulating cyclin-dependent kinase inhibitors. It promotes apoptotic pathways via activation of caspases and mitochondrial permeabilisation. At the signalling level, InsP6 disrupts the phosphoinositide 3-kinase/AKT/mTOR cascade and downstream mitogen-activated protein kinase pathways, attenuating growth factor-driven proliferation. Its metal-chelating properties diminish oxidative stress, reducing DNA damage and oncogenic transformation. InsP6 also modulates receptor–ligand interactions, inflammatory mediators and cytoskeletal architecture to impair migration and invasion. Preclinical studies in colorectal, prostate and breast tumour models have shown that InsP6, alone or combined with inositol, enhances chemotherapy efficacy, suppresses metastasis and improves quality of life. With a well-established safety profile, ongoing research centres on optimising formulation, dosing and delivery to translate these anticancer effects into clinical practice.
