Melano-Macrophage Centers in Fish Health Assessment
Summary
Melano-macrophage centres (MMCs) are aggregates of pigmented phagocytes primarily localised within the spleen and head kidney of teleost fish. These centres act as key sites for the sequestration of cellular debris, iron storage and antigen presentation, bridging innate immunity with emerging evidence for adaptive responses. Structurally reminiscent of mammalian germinal centres, MMCs accumulate melanin, lipofuscin and hemosiderin as by-products of erythrophagocytosis and oxidative processes. Changes in MMC size, number and composition have been correlated with stressors such as pollutant exposure, nutritional deficiencies, pathogen challenge and seasonal variations. Consequently, histological assessment of MMCs has gained traction as a practical, cost-effective biomarker for monitoring fish health in both wild and aquaculture settings. Advances in imaging and immunohistochemical staining have further refined the interpretation of MMC dynamics, underscoring their global significance for environmental monitoring and disease surveillance in increasingly impacted aquatic ecosystems.
