Metabolomic Approaches in Traditional Chinese Medicine

Summary

In recent years, metabolomic methodologies have been applied to Traditional Chinese Medicine (TCM) to bridge ancient holistic theory with modern systems biology. By profiling endogenous small molecules in biological samples, researchers can characterise the metabolic perturbations that underlie TCM syndromes and therapeutic interventions. Such approaches enable the identification of syndrome-specific biomarkers, the elucidation of active components within complex formulas and the elucidation of multi-target mechanisms. Innovations include the integration of serum pharmacochemistry, high-resolution mass spectrometry and multivariate statistical analysis to generate comprehensive metabolic maps. These strategies facilitate quality control, inform the standardisation of herbal preparations and accelerate the discovery of bioactive constituents. The global significance of this work lies in its potential to validate TCM efficacy, support regulatory approval and foster international collaboration in drug discovery. Practical applications range from the evaluation of processing methods that enhance therapeutic potency to the development of precision approaches for syndrome differentiation.

Building on foundational work in chinmedomics, an integrated strategy has been developed that combines serum pharmacochemistry with high-resolution metabolomics to discover the bioactive constituents of classic herbal formulas. By mapping in vivo parent compounds and metabolites, this platform identifies differential biomarkers and predicts molecular targets via network correlation models. In a case study of a kidney-yang deficiency remedy, dozens of signature peaks were characterised, leading to the prediction of key pathways linked to hormone regulation and energy metabolism. In parallel, high-throughput UPLC-MS analysis of Phellodendri Amurensis cortex applied chinmedomics to a prostate cancer model, revealing in vivo prototypes, metabolites and thirty-plus metabolic biomarkers that were normalised by ten absorbed constituents, which in turn were associated with anti-tumour targets. Further advancement of this strategy enabled the prediction of effective components in a multi-herb formula for kidney-yang deficiency, demonstrating multi-pathway regulation of lipid, steroid and amino acid metabolism and reinforcing the value of chinmedomics for target discovery.
