Summary
Oak decline represents a multifactorial syndrome in which the native microbiota of the tree shifts from a largely mutualistic community to a deleterious pathobiome. In healthy oaks, diverse bacterial and fungal assemblages inhabit the rhizosphere, phyllosphere and inner bark, contributing to nutrient cycling, defence priming and resilience to environmental stress. As decline progresses, specific bacteria—most notably Brenneria goodwinii, Gibbsiella quercinecans and Rahnella victoriana—proliferate within necrotic lesions in the inner bark, producing cell wall–degrading enzymes and other virulence factors that undermine host defences. Concurrently, interactions with the buprestid beetle Agrilus biguttatus exacerbate tissue damage and facilitate bacterial entry. Recent work has revealed that the emergence of the pathobiome is not solely a shift in species abundance but involves coordinated expression of virulence genes, modulation of reactive oxygen species by both host and microbes, and recruitment of secondary Gram-positive bacteria. These polymicrobial interactions operate within the broader holobiont framework, emphasising the dynamic dialogue between host physiology, microbial consortia and abiotic triggers such as drought. Understanding these processes at genomic, transcriptomic and ecological levels is essential to inform novel management strategies, including early detection of microbiome shifts, breeding for microbiome-mediated resistance and targeted microbiome engineering to restore oak health.
