Summary
Mössbauer spectroscopy has emerged as a powerful tool for probing the local environment of iron and other Mössbauer-active nuclei within biological systems. By exploiting the recoil-free resonance absorption of gamma rays, this technique provides precise measurements of hyperfine parameters—such as isomer shifts and quadrupole splittings—that reflect oxidation state, spin configuration and ligand coordination of iron centres. In biomedical investigations, Mössbauer spectroscopy has been applied to elucidate the structure and function of iron-containing proteins (including haemoglobin, myoglobin and ferritin), to monitor iron distribution in neural tissues, and to discern subtle changes in metal speciation associated with disease progression. Its sensitivity to sub-cellular iron pools enables discrimination between ferric and ferrous states, detection of iron–sulfur clusters in enzymes and characterisation of pathological iron deposits. Combined with complementary imaging modalities and micro-analytical methods, Mössbauer spectroscopy contributes to a more comprehensive understanding of iron metabolism in health and disease, guiding the development of diagnostic markers and therapeutic interventions for neurodegenerative disorders, anaemias and infections.
