Summary

Humor processing engages a distributed network that integrates cognitive appraisal, affective response and motor expression. Initial detection of an incongruity in a humorous stimulus recruits regions involved in attentional set‐shifting and semantic integration, notably the dorsolateral prefrontal cortex and middle temporal gyrus. Resolution of the incongruity often engages theory‐of‐mind and perspective‐taking regions such as the temporoparietal junction and precuneus. Successful resolution culminates in activation of mesocorticolimbic reward pathways—including the nucleus accumbens, amygdala and ventral tegmental area—where hedonic enjoyment is computed. Concurrently, motor readiness areas such as the supplementary motor area and premotor cortex prepare the facial and vocal components of laughter. This four‐stage cascade—expectation, incongruity detection, resolution and elaboration—provides a unified framework for understanding variations in humour style, developmental trajectories and individual differences. Beyond advancing basic neuroscience, these insights bear on clinical domains such as depressive disorders, autism spectrum conditions and social anxiety, where humour appreciation is often impaired. They also inform educational and therapeutic interventions that harness humour to enhance learning, social bonding and well‐being.

Gelotophobics show heightened activity in dorsal corticostriatal circuits but diminished activation in the ventral mesocorticolimbic network when processing both hostile and benign jokes, indicating increased top-down control and reduced bottom-up affective response.

Comparative investigations of humour and monetary rewards demonstrate a clear dissociation in neural processing. Monetary cues and outcomes predominantly activate nucleus accumbens–anterior cingulate circuits associated with reward anticipation and motivation, whereas humorous rewards evoke stronger amygdala–midbrain engagement during the enjoyment phase, underscoring a modality-specific hedonic pathway for social rewards.
