Neuroprotective Effects of Flavonoids in Cognitive and Neurodegenerative Disorders

Summary

Flavonoids are plant-derived polyphenolic compounds abundant in fruits, vegetables and beverages. They readily cross the blood–brain barrier and engage multiple cellular pathways implicated in neuronal survival and function. In preclinical systems, flavonoids such as quercetin, apigenin, luteolin and rutin have been shown to counteract hallmark features of neurodegenerative disorders, including amyloid-β aggregation, tau hyperphosphorylation, oxidative stress, neuroinflammation and synaptic dysfunction. These compounds activate antioxidant responses, inhibit pro-inflammatory signalling, promote autophagy and modulate neurotrophic factors, thereby preserving neuronal integrity and cognitive performance in animal and cellular models. Epidemiological data further suggest that habitual dietary intake of flavonoid-rich foods correlates with reduced risk of cognitive decline in ageing populations. By targeting convergent mechanisms of age-associated brain vulnerability, flavonoids offer promising lead structures for therapeutic development in Alzheimer’s disease, Parkinson’s disease and related disorders.

Recent research has demonstrated the neuroprotective capacity of apigenin in human induced pluripotent stem cell models of Alzheimer’s disease. Apigenin treatment attenuated calcium hyperactivity in diseased neurons, suppressed nitrosative stress and reduced caspase-dependent apoptosis. It also down-regulated pro-inflammatory cytokine release from both neurons and activated microglia, preserving neurite integrity under inflammatory challenge. These findings underscore the ability of a single flavonoid scaffold to modulate oxidative, inflammatory and apoptotic pathways in human neural cells, highlighting its potential as a lead compound for disease-modifying interventions.

Microglial activation: Transition of central nervous system immune cells into pro-inflammatory or reparative states in response to injury or disease.
