Summary

Oxidative stress arises from an imbalance between reactive oxygen species (ROS) and the body’s antioxidant defences, leading to potential damage to lipids, proteins and nucleic acids. In the oral cavity, saliva serves not only to lubricate and protect but also as a mirror of redox homeostasis. A spectrum of biomarkers measurable in saliva—including enzymatic antioxidants such as superoxide dismutase and non-enzymatic molecules such as uric acid—provides insight into local and systemic oxidative challenges. Markers of lipid peroxidation (for example malondialdehyde), protein carbonylation and DNA oxidation are elevated in common oral diseases such as dental caries, periodontitis and mucosal lesions, reflecting inflammation and microbial dysbiosis. Conversely, assays of total antioxidant capacity (TAC) and ferric reducing antioxidant power (FRAP) assess the cumulative protective potential of salivary constituents. The non-invasive nature of saliva collection, coupled with advances in sensitive analytical methods, has fuelled interest in redox indicators for early diagnosis, monitoring of therapeutic interventions and understanding links between oral and cardiovascular or metabolic disorders. Global research efforts aim to standardise sampling protocols and validate marker panels, with a view to translating salivary oxidative profiles into routine clinical practice and precision oral healthcare.

A recent investigation evaluated two formulations of sugar-free chewing gum enriched with spice-derived polyphenols and assessed their immediate impact on salivary redox status. Over a standard chewing interval, both products elicited significant increases in hydrophilic and lipophilic antioxidant capacity, alongside elevated phenolic content. This work demonstrates a practical strategy to bolster oral antioxidant defences and suggests potential for sublingual uptake of bioactive compounds. The findings pave the way for trials exploring whether sustained use of such functional gums can mitigate oral inflammation and influence systemic oxidative markers, thereby linking daily oral care routines with broader health outcomes.

Superoxide dismutase (SOD): An enzyme that catalyses the dismutation of superoxide radicals into oxygen and hydrogen peroxide, central to enzymatic antioxidant defence.
