Oxytocin Modulation of Social Behavior and Neuroendocrine Functions

Summary

Oxytocin is a hypothalamic nonapeptide that acts both as a neuromodulator in the brain and as a hormone in the periphery. Within the central nervous system, oxytocin contributes to social cognition by enhancing social recognition, trust, attachment and reducing anxiety in social contexts. It exerts these effects through activation of oxytocin receptors (Oxtr) located in key circuits including the hippocampus, amygdala and prefrontal cortex. Peripheral release of oxytocin coordinates critical neuroendocrine functions such as uterine contractions during parturition, milk ejection in lactation and modulation of the hypothalamic–pituitary–adrenal axis. Oxytocin is released in a pulsatile fashion, with amplitude and frequency of pulses adapting to physiological demands. Intranasal administration of oxytocin has become a widely used experimental tool to probe its central effects in humans, revealing influences on eye contact, emotion recognition and reward processing. Emerging work highlights the dual role of oxytocin in linking social behaviour to systemic physiological responses, suggesting therapeutic potential in disorders of social cognition, stress dysregulation and metabolic imbalance.

Recent studies have delineated a hippocampal pathway in which neuronal activity within the dorsal CA2 region projects to ventral CA1 to orchestrate distinct phases of social memory. Reversible inhibition of the dorsal CA2 impairs encoding, consolidation and recall of social encounters, demonstrating its dynamic importance across memory stages. Direct excitatory input from dorsal CA2 to a ventral CA1 subpopulation identified as social‐memory responsive reveals a sequential circuit essential for distinguishing conspecifics.

Complementary research has pinpointed oxytocin receptors in the anterior dentate gyrus and CA2/CA3 fields as necessary for discrimination of social stimuli. Genetic or pharmacological blockade of these receptors abolishes social but spares non‐social discrimination, while optogenetic mapping shows that anterior CA2/CA3 outputs to posterior CA1 specifically govern social recognition. Together, these findings establish a hippocampal Oxtr‐expressing axis that translates neuropeptide signalling into social memory computations.
