Summary
Timosaponins, a group of steroidal saponins predominantly isolated from Anemarrhena asphodeloides, have emerged as multifaceted agents in the fight against cancer. Among these, Timosaponin-AIII has attracted significant interest owing to its selective cytotoxicity towards tumour cells while sparing normal tissues. Mechanistically, timosaponins orchestrate a spectrum of anticancer actions: they induce apoptosis via mitochondrial disruption and endoplasmic reticulum stress, provoke cell-cycle arrest at G2/M through DNA damage signalling, modulate autophagy, and inhibit proliferative pathways such as PI3K/Akt and mTOR. Furthermore, they suppress tumour invasion and metastasis by targeting cytoskeletal dynamics and integrin/FAK signalling, and they exert anti-angiogenic and anti-inflammatory effects within the tumour microenvironment. Despite promising preclinical outcomes, the inherent hydrophobicity and limited bioavailability of timosaponins have prompted the development of advanced delivery systems to enhance their pharmacokinetic profiles. Collectively, these pharmacological activities underscore the potential of timosaponins as lead compounds for novel anticancer therapeutics.
