Pharmacological Applications of Nigella Sativa and Thymoquinone
Summary
Nigella sativa, commonly known as black cumin, has been employed in traditional remedies for centuries, with its seeds and oil valued for a broad spectrum of therapeutic properties. The principal bioactive constituent, thymoquinone, exhibits potent antioxidant, anti-inflammatory and immunomodulatory activities, which underpin its efficacy across varied pathological conditions. Preclinical investigations have demonstrated that thymoquinone modulates key signalling pathways—such as NF-κB, MAPK and PI3K/AKT—leading to attenuation of oxidative stress, suppression of pro-inflammatory cytokines and induction of apoptosis in malignant cells. In metabolic disorders, Nigella sativa seed extracts and oil improve glycaemic control, protect pancreatic β-cells and correct dyslipidaemia. Neuroprotective effects arise from mitigation of reactive oxygen species and enhancement of endogenous antioxidant enzymes, suggesting applications in neurodegenerative diseases. Anticancer studies reveal that thymoquinone exerts cytostatic and pro-apoptotic actions in diverse tumour models and can synergise with chemotherapeutic agents to overcome resistance.
