Summary

Diosgenin is a naturally occurring steroidal sapogenin found predominantly in species of Dioscorea, Trigonella and Smilax. It serves not only as a precursor for the synthesis of various steroidal drugs but also exhibits a broad spectrum of bioactivities relevant to cancer and chronic disease management. In oncology, diosgenin exerts anti-proliferative effects by inducing cell cycle arrest at G2/M or S phases, promoting mitochondrial-mediated apoptosis through Bcl-2 family modulation and activating caspase cascades. It also attenuates metastatic potential via suppression of matrix metalloproteinases and inhibition of key signalling axes such as PI3K/Akt, MAPK and NF-κB. Beyond its anticancer profile, diosgenin demonstrates anti-inflammatory and antioxidant actions that underpin its efficacy in metabolic disorders, atherosclerosis and neurodegeneration.
