Summary
Eurycoma longifolia, commonly known as tongkat ali, is a tropical medicinal plant renowned for its rich array of phytochemicals, notably quassinoids, alkaloids and phenolic compounds. Bioactive quassinoids such as eurycomanone and eurycomalactone exhibit androgen‐modulating, anti‐inflammatory and antioxidant properties. These compounds act on steroidogenic pathways to enhance testosterone synthesis, support spermatogenesis and influence signalling cascades including NF-κB and MAPK. In preclinical models they promote osteoblast proliferation, inhibit osteoclast activity and protect against oxidative stress, thereby addressing androgen deficiency–related bone loss. Standardised extracts have been shown to improve libido, erectile function, semen quality and stress hormone profiles in men. Advances in analytical techniques have also enabled metabolomic fingerprinting to ensure product authenticity and consistency. Together, these findings underpin the global significance of E. longifolia as a complementary approach to male reproductive health, stress management and bone preservation.

Recent studies have applied an integrated metabolomics approach to authenticate and standardise tongkat ali products. Nuclear magnetic resonance and gas chromatography–mass spectrometry analyses identified a suite of primary metabolites, quassinoids and volatiles that distinguish authentic root extracts from commercial formulations. Multivariate analysis, including principal component analysis and orthogonal partial least squares–discriminant analysis, revealed that specific quassinoids and fatty acids are enriched in genuine material. Aroma profiling further differentiated authentic samples by the presence of compounds such as E-cinnamaldehyde and terpinen-4-ol, whereas vanillin and maltol predominated in blends. Such fingerprinting ensures quality control and safeguards consumers against adulteration.
