Summary
Proteomic analysis of synovial fluid has emerged as a powerful approach to decipher the molecular underpinnings of joint degeneration. Synovial fluid bathes articular surfaces and reflects the local biochemical milieu of cartilage, synovium and subchondral bone. By applying advanced mass spectrometry and fractionation strategies, researchers have catalogued hundreds of proteins, including glycoproteins, cytokines and extracellular matrix fragments. Comparative profiling between healthy joints and those afflicted by osteoarthritis or rheumatoid arthritis has revealed disease-specific signatures, highlighted dysregulated pathways of cartilage breakdown and inflammation, and identified neopeptides deriving from matrix proteolysis. Such discoveries underpin efforts to develop early diagnostic biomarkers, stratify patients for targeted therapies and monitor therapeutic response. Advances in sample preparation, peptide‐based depletion of high‐abundance proteins and label-free quantification have enhanced depth of coverage and reproducibility.
