Quorum Sensing Mechanisms in Staphylococcus aureus Pathogenesis

Summary

Staphylococcus aureus employs a sophisticated quorum sensing network to coordinate virulence, biofilm formation and dispersal in response to population density. Central to this network is the accessory gene regulator (agr) system, a peptide-mediated two-component circuit that controls expression of toxins, enzymes and adhesion factors. At low cell density, agr activity is minimal, favouring surface attachment and biofilm maturation. As cell numbers rise, accumulation of an autoinducing peptide (AIP) ligand triggers a sensor kinase, AgrC, which phosphorylates the response regulator AgrA. Activated AgrA induces RNAIII transcription, upregulating exoproteins and downregulating surface adhesins. This switch orchestrates biofilm detachment, immune evasion and tissue invasion. Cross-talk among agr subtypes and interference by peptides from other microbes further modulates S. aureus behaviour in mixed communities. Quorum sensing thus underpins the balance between chronic colonisation and acute infection, making agr signalling a pivotal target for novel anti-virulence strategies.

Quorum Sensing Mechanisms in Staphylococcus aureus Pathogenesis publication trend

The graph below shows the total number of articles in quorum sensing mechanisms in staphylococcus aureus pathogenesis across all publications each year (not limited to Nature Index journals).

Technical terms

Quorum Sensing: Cell-density dependent communication by secreted signals that synchronise gene expression across a bacterial population.

Autoinducing Peptide (AIP): Short, post-translationally modified peptide that accumulates extracellularly to activate a cognate sensor kinase in Gram-positive bacteria.

Accessory Gene Regulator (agr) System: A peptide-mediated two-component regulatory circuit that controls virulence gene expression and biofilm dynamics in S. aureus.

Biofilm: Structured bacterial community embedded in an extracellular matrix, providing protection from antimicrobials and host defences.

Quorum Quenching: Interference with quorum sensing signalling to attenuate virulence without killing the pathogen.

Virulence Factor: Molecule or structure produced by a pathogen that contributes to disease causation or severity.

About these summaries

This Nature Research Intelligence Topic summary is created with the cited references and a large language model. We take care to ground generated text with facts, and have systems in place to gain human feedback on the overall quality of the process in line with our AI principles. We strive to create accurate and useful summaries for people unfamiliar with the research topic and that supports this goal. These pages are a beta release and will be updated as we learn how best to help people gain value from a research topic summary.
